Deciphering Scaffolding Roles on non-coding RNAs in Protein Condensation

We will explore the integration of computational and experimental strategies to unravel the complex interaction webs involving XIST [1] and RNAs associated with neurodevelopmental disorders such as Fragile X-associated Tremor/Ataxia Syndrome (FXTAS) [2], as well as neurodegenerative diseases, including Parkinson’s and Amyotrophic Lateral Sclerosis (ALS) [3]. Our examination centers on the mechanisms that facilitate harmful liquid-to-solid phase transitions within cells. Highlighting the seminal discovery that non-coding RNAs function as scaffolds for protein-RNA interactions [4], our research has spotlighted specific transcript attributes critical for forming extensive RNA-protein complexes and instigating phase separation [5]. This knowledge is fundamental for comprehending cellular behaviors, especially regarding viral infection mechanisms [6]. Most importantly, our advancements in engineering synthetic RNAs to modify the condensation characteristics of proteins implicated in ALS represent a pivotal progression in our efforts to mitigate these debilitating conditions [7]. New data on RNA aptamers will be discussed.
